Once the semen dries out the sperm should be dead. So if the wet spot is no longer wet then the sperm should all be dead. If you want to make sure the sperm are off your penis go lather up real quick with some soap and rinse them away. Inside the woman it has been suggested that they may live up to seven days, but usually many die by that time. The best chance of pregnancy is about 1-3 days before ovulation, so I expect that after 3-4 days most sperm inside the body die, but now I'm just rambling. To answer your question: once the semen dries out the sperm should be dead. If you would like to be sure they are all gone go wash your penis. Additionally, since some my still be present in your urethra(the tube through which sperm are delivered and you go to the bathroom) then going to the bathroom should remove all the sperm that remain inside your penis as well.
