I hope this is the right forum to post this.
My wife goes for her first mammogram in about two weeks. What can she expect as far as the amount of pain she will endure? What can I do to comfort her?

It's not painful, it's uncomfortable. It's no worse then having a pap done. As far as supporting her, not much to do unless there is an abnormal finding. It's really no big deal, you just get your breast compressed for a few minutes and then you are on your way.
